A protein sample is mixed with the 2X sample buffer (1:1) and heated in boiling water for 2-5 min. The DTT reduces the intra and inter-molecular disulfide bonds. The SDS detergent denatures the proteins and subunits and gives each an overall negative charge so that each will separate based on size. The bromophenol blue serves as a dye front that runs ahead of the proteins and also serves to make it easier to see the sample during loading.
