描述：

Tiamulin is a semisynthetic pleuromutilin antibiotic that binds to the ribosomal peptidyl transferase centre and inhibits protein synthesis.

体外研究：

Tiamulin has a remarkable effect on decreasing the activity of CD73 in vitro and dramatically inhibits colony formation of MDA-MB-231 and 4 T1 cells. Tiamulin is located within the peptidyl transferase center (PTC) of the 50S ribosomal subunit with its tricyclic mutilin core positioned in a tight pocket at the A-tRNA binding site. Tiamulin directly inhibits peptide bond formation. It interacts with 23S rRNA and clearly interferes with the correct positioning of both A- and P-site substrates.

体内研究：Tiamulin inhibits breast tumor growth and metastasis, mainly achieved by affecting the activity but not expression of CD73. The antitumor effect of Tiamulin results in inhibition of metastasis and angiogenesis.
