Background
This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- C5a antibody was pre-coated onto 96-well plates. And the biotin conjugated anti-C5a antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP-Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blu e color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the C5a amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of C5a can be calculated. C5a is a protein fragment released from cleavage complement component C5 by protease C5-convertase into C5a and C5b fragments. C5b is important in late events of complement cascade, whereas C5a acts as highly inflammatory peptide. The origin of C5 is in the hepatocyte but its synthesis can also be found in macrophages that may cause local increase of C5a. C5a has chemotactic and anaphylatoxic properties, it is essential in the innate immunity but it is also linked with the adaptive immunity.
