传代培养及其操作步骤:原代细胞培养成功以后,需要进行分离培养,否则细胞会因生存空间不足或密度过大,营养障碍,影响细胞生长。细胞由原培养瓶内分离稀释后传到新的培养瓶中培养的过程称之为传代培养。传代细胞允许培养的细胞扩增(形成细胞株),可以进行细胞克隆,易于保存,但可能丧失一些特殊的细胞和分化特征。传代细胞形成细胞株的利处在于提供了大量持久的实验材料,便于实验。传代细胞培养操作步骤:1)吸掉或倒掉培养瓶内旧培养液;2)向瓶内加入胰蛋白酶液和EDTA混合液少量。以能覆盖培养瓶底为宜;3)置37℃孵箱或室温(25℃温度)下进行消化,2-5min后把培养瓶放在倒置显微镜下进行观察,当发现胞质回缩、细胞间隙增大后,应立即中止消化;4)吸出消化液,向瓶内加入Hanks液少量,轻轻转动培养瓶,把残留消化液冲掉,然后再加培养液。如果仅使用胰蛋白酶消化,在吸除胰蛋白液后,可直接加入少量含血清的培养液,终止消化;5)使用弯头吸管,吸取瓶内培养液,按顺序反复轻轻吹打瓶壁细胞,使之从瓶壁脱离形成细胞悬液。吹打时动作要轻柔,以防用力过猛损伤细胞;6)用计数板计数后,分别接种于新的培养瓶中,置CO2培养箱中进行培养;7)细胞培养换液时间应根据细胞生长的状态和实验要求来确定。一般2-3d后应换一次生长液。待细胞铺满器皿底面,即可使用;也可继续传代扩大培养或换成维持液。

细胞冻存的步骤:1)选择处于对数生长期的细胞,在冻存前一天Z好换液。将多个培养瓶中的细胞培养液去掉,用0.25% 胰蛋白酶消化。适时去掉胰蛋白酶,加入少量新培养液。用吸管吸取培养液反复吹打瓶壁上的细胞,使其成为均匀分散的细胞悬液。悬浮生产细胞则不要消化处理。然后将细胞收集于离心管中离心(1000r/min,10分钟);2)去上清液,加入含20%小牛血清的完全培养基,于4℃预冷15分钟后,逐滴加入已无菌的DMSO或甘油,用吸管轻轻吹打使细胞均匀,细胞浓度为5×106~1×107/mL之间。(冻存培养液的配置是不是一定要用小牛血清呢?答案是不一定的,具体要看培养的细胞类型来选择;3)将分装上述细胞悬液于2ml冻存管中,每管0.25ml。冻存管要将盖子盖紧,并标记好细胞名称和冻存日期,同时作好登记(日期、细胞种类及代次、冻存支数);4)将装好细胞的冻存管放到冻存盒中,-80℃冰箱过夜。;如果有程序降温器(放在-80℃冰箱过夜,放入液氮罐)Z好;或者可以在4℃,2h;然后转到-20℃,2h;-80℃,2h;放入液氮罐;5)细胞冻存在液氮中可以长期保存,但为妥善起见,冻存半年后,Z好取出一只冻存管细胞复苏培养,观察生长情况,然后再继续冻存。
