Cadherin-8 (CDH8) is a type II classical cadherin from the cadherin superfamily. Member of the Cadherin superfamily are integral membrane proteins that mediate calcium-dependent cell-cell adhesion. Cadherin proteins are composed of a large N-terminal extracellular domain, a single membrane-spanning domain, and a small highly conserved C-terminal cytoplasmic domain. Cadherins are calcium dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner in connecting cells. The extracellular domain of CDH8 contains five cadherin domains. CDH8 is expressed in brain and is putatively involved in synaptic adhesion, axon outgrowth and guidance.

CD40 is a Type I Transmembrane Glycoprotein that belongs to the TNF Receptor Superfamily. CD40 is expressed in B cells, follicular dendritic cells, dendritic cells, activated monocytes, macrophages, endothelial cells, vascular smooth muscle cells, and several tumor cell lines. The extracellular domain of CD40 is characterized by Cysteine rich repeat regions. Interaction of CD40 with its ligand (CD40L) leads to aggregation of CD40 molecules, which in turn interact with cytoplasmic components to initiate signaling pathways. Several different TRAF proteins (adaptor proteins) have been identified to serves as mediators of the signal transduction. CD40 plays an essential role in mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.

Complement is defined as key part of innate immunity and as the first line of defense in the fight against invading pathogens. Complement 3 (C3) is the most abundant component of the complement cascade and the convergent point for all three major complement activation pathways: namely classical, alternative and mannose-binding lectin pathways. Complement activation leads to the formation of the C3 convertase, which cleaves C3 into the key effector molecules, C3a (anaphylatoxin) and C3b (opsonin) which then drive microbe removal. By binding to C3a receptor (C3aR), C3a exhibits potent anaphylatoxin activity, including increased vascular permeability, triggering degranulation of mast cells, inflammation, and activating leukocytes.
