D-荧光素钾（D-luciferin potassium）

Description/产品描述
D-Luciferin 作为荧光素酶的底物，存在于多种发光生物体中。在ATP和荧光素酶的催化作用下，Luciferin被氧化，产生蓝绿色的光（560nm），当底物过量时，产生的光量子数与荧光素酶的浓度呈正相关。编码荧光素酶的Luc基因是植物、细菌、哺乳动物细胞的常用报告基因。由于没有背景干扰，因此可以很容易地检测出低至 0.02 pg水平的荧光素酶。

Protocol for In vivo analysis / 活体成像分析方法 
1．用DPBS（w/o Mg2+、Ca2+）配置D-luciferin工作液（15mg/mL）* ，0.2 um滤膜无菌过滤。 
2．注射量：10μL/g的体重，如10 g重小鼠，注射100μL储备液（1.5 mg Luciferin）。 
3．腹腔注射 （i.p.） 10-15分钟后，上机进行图像分析。

Protocol for In vitro analysis / 体外分析方法 
1. 用无菌水配置200×储备液（30mg/ml）*，立即使用，或-20℃保存。
2. 用预热好的完全培养基1∶200稀释Luciferin储备液，配置工作液（150μg/mL）。 
3. 去除培养细胞的培养基。 
4. 进行图像分析前，向细胞中添加1×的Luciferin工作液，进行图像分析**。

* ：溶解D-luciferin时要完全。 
**：在图像分析前，将细胞进行37℃的短时间培养可以增加信号强度。
