N-hydroxysuccinimidyl esters (NHS) of biotins are excellent reagents for biotinylating biomolecules such as proteins and amine-modified nucleic acids. NHS-Biotin (Order-No. B001N) is the simplest biotinylating reagent available. When the target amines are less reactive (i.e. secondary amines) a fluorophenol active ester like PFP-Biotin (Order-No. B010F) is preferred. Both reagents are insoluble in aqueous media, and must be first dissolved in non-protic organic solvents such as DMF or DMSO, then aliquot into the aqueous reaction mixture. Because these compounds are prone to hydrolysis by water, it is recommended to prepare fresh stock solutions to achieve the best yield at a final concentration of organic solvent not above 10%. These active esters contain a negatively charged sulfonate in their NHS ring structure. They can be added directly to aqueous reaction media, without the need of organic solvents. Concentrated stock solutions may be prepared, but must used immediately as they hydrolyze quickly.  Another water-soluble Sulfo NHS ester like Sulfo NHS-SS-Biotin (Order-No. B104N) contains a spacer arm which can be cleaved under mild conditions. Reduction of the disulfide bond with 50 mM dithiothreitol releases the biotinylated component from the avidine or streptavidine labeled detection reagents. Cyanagen developed a new class of water-soluble Biotin NHS esters based on hydrophilic polyethylene oxide (PEO) spacer arms like:
