Fluo-3 absorbs at 506 nm and emits at 526 nm when bound to calcium. It is one of the most widely used fluorescent indicators because it can be optimally excited by the commonlyused 488 argon-ion laser line and its relatively long wavelength emission reduces interference from background fluorescence. Unlike Fura-2 and Indo-1, neither the excitation nor the emission maximum of the dye shifts significantly before and after Ca2+ binding. As a result, the fluorometric ratioing technique is not applicable to Fluo-3. Fluo-3 is essentially nonfluorescent without Ca2+ present, but the fluorescence increases at least 40 times on Ca2+ binding. Because Fluo-3 binds Ca2+ more weakly (higher Kd) than Fura-2 and Indo-1, it is more useful for measuring high transient Ca2+ concentration during Ca2+ spikes.
Like other AM esters, Fluo-3 AM ester is relatively unstable when frequently exposed to moisture. For this reason, Biotium offers Fluo-3 AM ester in several packaging sizes including 20x50µg and 10x100µg, which facilitate storage and handling. Fura-2 is a widely used UV-excitable fluorescent calcium indicator.1-5 Upon calcium binding, the fluorescent excitation maximum of the indicator undergoes a blue shift from 363 nm (Ca2+-free) to 335 nm (Ca2+-saturated), while the fluorescence emission maximum is relatively unchanged at ~510 nm. The indicator is typically excited at 340 nm and 380 nm respectively and the ratio of the fluorescent intensities corresponding to the two excitations is used in calculating the intracellular concentrations. Measurement of calcium concentration using this ratioing method avoids interference due to uneven dye distribution and photobleaching (Bright, G.R., et al, in Fluorescence Microscopy of Living Cells in Culture, Part B, (Methods in Cell Biology, Vol. 30), Academic Press (1989) p. 157). Fura-2 has been used in many cellular systems and applications, particularly in microscopic imaging.
Furaptra, which is also commonly called Mag-Fura-2, was originally developed as a fluorescent magnesium indicator. However, it is an excellent indicator for high calcium in the range of 1µM to 100 µM.6,7 Fura-2 and Furaptra share the same chromophore and as a result have similar spectral change in response to calcium concentration change.
