As a a potent small-molecule inhibitor of antiapoptotic proteins, ABT-737 can inhibit Bcl-2, Bcl-w and Bcl-xl with EC50 of 30.3 nM, 78.7 nM and 198.7 nM.
For cell lines
(1)Acute myeloid leukemia
ABT-737 effectively kills acute myeloid leukemia blast, progenitor, and stem cells without affecting normal hematopoietic cells. ABT-737 induced the disruption of the BCL-2/BAX complex and BAK-dependent but BIM-independent activation of the intrinsic apoptotic pathway. In cells with phosphorylated BCL-2 or increased MCL-1, ABT-737 was inactive. Inhibition of BCL-2 phosphorylation and reduction of MCL-1 expression restored sensitivity to ABT-737.
(2)Human MM cells
ABT-737 induces apoptosis in MM cells, including those resistant to conventional therapy. ABT-737 decreases the viability of bortezomib-, dexamethasone-(Dex) and thalidomide-refractory patient MM cells. ABT-737 abrogates MM cell growth triggered by interleukin-6 or insulin-like growth factor-1. ABT-737-induced apoptosis is associated with activation of caspase-8, caspase-9 and caspase-3, followed by poly(ADP-ribose) polymerase cleavage.
