重组人TXN蛋白 Recombinant Human Thioredoxin/TXN/Trx

细胞因子检测细胞因子的检测有助于了解其在免疫调节中的作用, 鉴定分离的淋巴细胞, 监测某些疾病状态的细胞免疫功能。例如, 根据培养的 CD4+T 细胞分泌的细胞因子确定细胞亚群, 产生 IL - 12 和 IFN -γ者为 Th1 细胞, 产生 IL - 4 和 IL - 10 者为 Th2 细胞; 艾滋病患者 IL - 2 水平明显降低, 而类风湿性关节炎、多发性硬化、移植排斥反应等患者的 IL - 2水平则升高。细胞因子的检测方法可分以下 3 种: (1) ELISA。用细胞因子单克隆抗体包被固相的双抗体夹心法或 ELISPOT 测定法检测细胞因子, 例如检测 IL - 2, IL - 5, IL - 6。亦可用荧光素标记细胞因子抗体, 对胞内细胞因子进行染色, 直接以流式细胞仪测定产生该细胞因子的细胞数目。 (2) 生物活性测定法。可根据细胞因子的生物活性, 选用相应的实验系统, 包括细胞增殖法、直接杀伤法、保护细胞免受病毒致病变法等。如选用细胞因子依赖的细胞株, 其增殖反应与细胞因子的量成正相关, 根据细胞株的增殖水平可确定样品中细胞因子的含量, 如检测 IL - 1, IL - 2, IL - 4, IL - 6。 (3) 聚合酶链反应 (PCR)。根据细胞因子的核苷酸序列, 设计特定细胞因子的引物, 利用逆转录 PCR 测定待检细胞中特异的 mRNA。该法可用于多种细胞因子的测定。
