本品是含有蓝色和橙色染料(电泳位置分别约4kb和50bp)的2倍浓度的PCR预混液,含有2X Taq DNA Polymerase、2X PCR Buffer、2X dNTP和2X Loading Buffer,只需加入模板DNA、引物和水即可进行PCR扩增,并且扩增完毕后可以直接上样电泳。主要适用于cDNA或基因组DNA等的常规PCR扩增,特别适合于定性或半定量的PCR扩增,也可用于长度小于2kb的DNA*(代'片')段的扩增和克隆。
本品中已经含有所有的通用组分,用户只需自备引物、模板DNA和水即可进行PCR扩增。大大简化了PCR操作,使操作更加快捷,也减少了PCR操作过程中可能导致的污染,使PCR的重复性更好。可以扩增出长达8kb的片段,但通常更适合用于扩增2kb以下的DNA*(代'片')段。
本产品中加入了蓝色和橙色的电泳示踪染料(整体呈现绿色),其在浓度为1%琼脂糖胶中的迁移位置分别大约位于4kb和50bp处。PCR结束后可直接进行电泳检测,无需再添加上样缓冲液。蓝色和橙色示踪染料不会影响对相应DNA条带的观察和检测。
本产品稳定性高,经测试,反复冻融15次后对PCR的扩增效果无显著影响。反复冻融15次前后,使用不同引物扩增100-1500bp间不同长度目的片段的效果如下图所示。

Background | Hsp27 is a chaperone of the sHsp (small heat shock protein) group among ubiquitin, α-crystallin, Hsp20 and others. The common functions of sHsps are chaperone activity, thermotolerance, inhibition of apoptosis, regulation of cell development, and cell differentiation. They also take part in signal transduction. Hsp27 appears in many cell types, especially all types of muscle cells. It is located mainly in the cytosol, but also in the perinuclear region, endoplasmatic reticulum, and nucleus. It is overexpressed during different stages of cell differentiation and development. This suggests an essential role for Hsp27 in the differentiation of tissues. Probably Hsp27 – among other chaperones – is involved in the process of cell differentiation. Changes of Hsp27 levels were observed in Ehrlich ascite cells, embryonic stem cells, normal B-cells, B-lymphoma cells, osteoblasts, keratinocytes, neurons etc. |
