    pET-19b Description

    pET-19b plasmid is a prokaryotic expression vector with a 10 x His label and a BamHI restriction endonuclease site in the N terminal. The expression is induced by the T7 RNA polymerase provided by the host cell. The target gene is cloned to plasmid vector and controlled by the strong transcription and translation signal of phage.


    pET-19b plasmid is the most powerful system in pET system which has ever been able to express the recombinant protein in Escherichia coli and is the most widely used system for prokaryotic expression. This series of plasmids can easily weaken the protein expression by reducing the concentration of the inducer. Under non induction conditions, the target gene can be completely silent and not transcribed.

    pET-19b vector carries an N-terminal His•Tag® sequence followed by an enterokinase site and three cloning sites. Unique sites are shown on the circle map. Note that the sequence is numbered by the pBR322 convention, so the T7 expression region is reversed on the circular map. The cloning/expression region of the coding strand transcribed by T7 RNA polymeraseis shown below.
