Background
 
This kit was based on sandwich enzyme-linked immune-sorbent assay technology. Anti- CERKantibody was pre-coated onto 96-well plates. And the biotin conjugated anti- CERK antibody was used as detection antibodies. The standards, test samples and biotin conjugated detection antibody were added to the wells subsequently, and wash with wash buffer. HRP- Streptavidin was added and unbound conjugates were washed away with wash buffer. TMB substrates were used to visualize HRP enzymatic reaction. TMB was catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the CERK amount of sample captured in plate. Read the O.D. absorbance at 450nm in a microplate reader, and then the concentration of CERK can be calculated.

About Ceramide Kinase / CERK
 
CERK, which phosphorylates Cer to produce ceramide 1-phosphate (C1P), was initially described as a Ca2+-stimulated lipid kinase that copurified with brain synaptic vesicles. CERK activity has been reported in HL60 cells, mast cells, and neutrophils. CERK is involved in phagolysosome formation in polymorphonuclear leukocytes and also in liposome fusion. C1P has been reported to have mitogenic effects. More recently, C1P was found to induce arachidonic acid release and prostanoid synthesis. Uniprot: Q8TCT0 NCBI: NM_022766 NP_073603.2
