IMUBIND® ADAMTS13 Autoantibody ELISA（血管性血友病因子裂解酶自身抗体ADAMTS-13Ab酶联免疫试剂盒）

 INTENDED USE
The IMUBIND® ADAMTS13 Autobody ELISA is intended for the measurement of ADAMTS13 IgG autoantibodies in human plasma. The assay is limited to “For Research Use Only” in the United States.

PRINCIPLE OF THE METHOD
Diluted plasma samples are added to microwells coated with a full-length recombinant ADAMTS13. During an incubation period, autoantibodies to ADAMTS13 present in the sample will bind to the protein coated to the wells. Following a washing step, a goat anti-human IgG antibody labeled with horseradish peroxidase (HRP) is added to the microwells and binds to IgG type ADAMTS13 autoantibodies that have bound to the coated microwells. Following another washing step, the addition of a perborate-3,3’-5,5’-tetramethylbenzidine (TMB) substrate and its subsequent reaction with the HRP present generates a blue colored solution. The reaction is stopped by adding sulfuric acid, which turns the solution yellow. Measuring the solution absorbance at 450 nm and comparing the value with those of a standard curve determines the level of ADAMTS13 autoantibodies in the diluted plasma sample. ADAMTS13 autoantibody levels are expressed in Arbitrary Units/mL (AU/mL). One AU/mL is equal to 1 μg/mL of affinity purified human anti-ADAMTS13 IgG.
