INTENDED USE
The IMUCLONE® Rat PAI-1 ELISA is an enzyme-linked immunosorbent assay for the measurement of rat plasminogen activator inhibitor type 1 (PAI-1) in plasma, biological fluid or cell culture supernatant. The assay reacts with latent, active and inactive PAI-1 as well as with tPA/PAI-1 complexes. This assay is for investigational use in laboratory animals or other tests that do not involve human subjects.

PRINCIPLE OF THE METHOD
The IMUCLONE Rat PAI-1 ELISA is a sandwich ELISA utilizing a pair of monoclonal antibodies specific for rat PAI-1 reacting with two different, yet complementary epitopes. First, the Detection Antibody, a mouse monoclonal antibody against rat PAI-1 coupled to horseradish peroxidase (HRP), is added to a microwell coated with a second mouse monoclonal antibody recognizing a different epitope of rat PAI-1. This is followed by the addition of diluted plasma or biological fluid to the microwell, where the Detection Antibody/PAI-1 complex is captured to the solid phase by the coating antibody against PAI-1. After the microwells are washed the peroxidase substrate, 3,3',5,5'–Tetramethylbenzidine (TMB), is added to the microwell. The subsequent enzyme reaction yields a blue colored solution. Addition of sulfuric acid stops the reaction and turns the solution color to yellow. The amount of color (i.e. the absorbance of the solution at 450 nm) is directly proportional to the amount of PAI-1 present in the sample.
