Description
Amphotericin B is a polyene antifungal.
TOKU-E offers three forms of amphotericin B: amphotericin B, USP (A007), amphotericin B, EP (A064), and amphotericin B, solubilized (A008). Amphotericin B, USP and amphotericin B, EP are nearly insoluble in aqueous solution (0.750 mg/mL). Amphotericin B, solubilized is comprised of 45% amphotericin B, 35% sodium deoxycholate, 10% phosphate buffer.
Mechanism
Amphotericin B targets ergosterol in the fungal cell membrane and increases permeability. This alteration of the cell membrane is toxic to the fungus and leads to death.
Spectrum
Amphotericin B targets a wide range of susceptible fungi including Histoplasma, Coccidioides, and Cryptococcusspecies.
Microbiology Applications
Amphotericin B is commonly used in clinical in vitro microbiological antimicrobial susceptibility tests (panels, discs, and MIC strips) against a wide range of fungal specimens. Medical microbiologists use AST results to recommend antifungal treatment options for infected patients. Representative MIC values include:
Cryptococcus neoformans 0.625 μg/mL – 2.5 μg/mL
Histoplasma capsulatum 0.03 μg/mL - 2 μg/mL
